zebrafish dickkopf 1 (dkk1) ORF fused to GFP (mmGFP5) at it's C-terminus. in pCS2P+ for expression from CMV promoter or in vitro sense RNA synthesis wirh SV40 polyA 3'UTR. dkk1 ORF amplified from 144 using primers 230+231, cloned PacI / AscI into 160. injection of 180ng/ul of this RNA produces VERY strong Wnt LOF phenotype (large heads, no posterior development), also GFP bright, albeit very diffuse (secreted protein?).
