Markerless Insertion Procedure

pBlPrt is a plasmid that allows you to make markerless insertions into the upt site of M. maripaludis. You can follow the same steps as listed in the protocol for pCRPrtNeo (markerless mutations), or use an alternative method that has been established in order to get your strain in as little as six days. This alternative method eliminates growth on plates until the final step.
