The characterization and control of solid/liquid interfaces is at the forefront of a range of topics including regenerative medicine, biofouling, implant technology, and medical diagnostics. My primary research activities originate from the need to probe conformation and orientation of biomolecules (DNA, peptides, proteins, antibodies...) in contact to a solid surface with molecular-level resolution and under standard conditions in situ and in vitro. My group has developed and applied femtosecond based sum-frequency-generation (SFG) spectrometers and several measuring cells including a thin layer analysis and a spectro-electrochemical cell. These have been used to study model systems such as self-assembled monolayers (SAMs) with various functional groups in air and liquid environments under controlled temperature, solution, and potential conditions. The information obtained by these experiments facilitated our understanding of SFG signals from more complex biomolecules such as DNA and peptides. Other systems of interest are supported lipid bilayers, membrane proteins, and stimuli-responsive polymer coatings. Recent work from my group has shown that SFG spectroscopy can be used to interrogate ordering phenomena within the extracellular matrix beneath adherent cells on artificial substrates at early stages of cell adhesion. The groundwork presented in these studies opens the possibility for use of this technique in a variety of other systems including the growth and development of biofilms or dynamic processes occurring during cell adhesion on functional surfaces.
